DAB Chromogenic Kit for Life Science Research

I. Product Introduction

The DAB Chromogenic Kit is designed for chromogenic detection in immunohistochemistry (IHC), in situ hybridization (ISH), or Western blot applications utilizing horseradish peroxidase (HRP)-labeled samples. DAB serves as a substrate for HRP, catalyzing the formation of an insoluble brown precipitate that is insoluble in both water and ethanol. This property allows subsequent staining with alcohol-soluble dyes after DAB development. For IHC or ISH applications, using 50 μL of working solution per sample, this kit supports detection for a minimum of 200 samples.

Transport: Wet ice.

Storage: Protect from light at 2–8°C. Shelf life: 12 months.

IV. Procedure (Please read the precautions before starting the experiment):

1. Preparation of DAB Working Solution:

Add 20 μL of 50× DAB Stock to every 1 mL of DAB Diluent and mix thoroughly. Prepare fresh immediately before use and use within the same day; long-term storage is not recommended.

2. Staining Procedure:

Apply the DAB working solution directly onto prepared IHC sections or Western blot membranes. Incubate at room temperature in the dark for 3–30 minutes or longer, until the desired color intensity is achieved.

3. Termination:

Remove the DAB working solution and wash twice with PBS or distilled water to stop the reaction.

V. Notice：

If excessive background staining occurs in IHC, consider extending wash times, using appropriate blocking solutions, quenching endogenous peroxidase activity, reducing incubation time, or lowering the secondary antibody concentration.

If weak or no signal is observed, increase primary or secondary antibody concentrations, extend development time, or verify secondary antibody activity.

DAB is harmful to humans. Handle with care, avoid direct contact or inhalation, and ensure proper protective measures are in place.

Always wear laboratory coats and disposable gloves during operation.
